چکیده

Background: Activation loop Y807F or juxtamembrane domain Y559F mutations compromise CSF-1 receptor-mediated macrophage proliferation. Results: Tyr-559 suppresses constitutive proliferative activity of Tyr-807; ligand-induced Tyr559 phosphorylation relieves this inhibition and activates pathways. Conclusion: Tyr-807 drives proliferation. Tyr-559 confers ligand dependence. Significance: How individual CSF-1 receptor tyrosines regulate receptor activation and signaling is critical for understanding the function of this disease-relevant receptor.

Colony-stimulating factor-1 blockade by antisense oligonucleotides and small interfering RNAs suppresses growth of human mammary tumor xenografts in mice.

Colony-stimulating factor (CSF)-1 is the primary regulator of tissue macrophage production. CSF-1 expression is correlated with poor prognosis in breast cancer and is believed to enhance mammary tumor progression and metastasis through the recruitment and regulation of tumor-associated macrophages. Macrophages produce matrix metalloproteases (MMPs) and vascular endothelial growth factor, which ...

The Role of Atypical Protein Kinase C in CSF-1-Dependent Erk Activation and Proliferation in Myeloid Progenitors and Macrophages

Colony stimulating factor-1 (CSF-1 or M-CSF) is the major physiological regulator of the proliferation, differentiation and survival of cells of the mononuclear phagocyte lineage. CSF-1 binds to a receptor tyrosine kinase, the CSF-1 receptor (CSF-1R). Multiple pathways are activated downstream of the CSF-1R; however, it is not clear which pathways regulate proliferation and survival. Here, we i...

Phosphorylation of CSF-1R Y721 mediates its association with PI3K to regulate macrophage motility and enhancement of tumor cell invasion.

Colony stimulating factor-1 (CSF-1) regulates macrophage morphology and motility, as well as mononuclear phagocytic cell proliferation and differentiation. The CSF-1 receptor (CSF-1R) transduces these pleiotropic signals through autophosphorylation of eight intracellular tyrosine residues. We have used a novel bone-marrow-derived macrophage cell line system to examine specific signaling pathway...

P-76: Important Role of Macrophage Colony-Stimulating Factor during Menstrual Cycle and Early Pregnancy Phase in Oovarian Stimulated IVF-Ppatients

Background: M-CSF is a cytokine mediating the growth, proliferation and differentiation of various cell types including macrophages, trophoblast cells, and osteoclasts. It plays an important role in immunity reproduction, follicle development and ovulation. In the present study, we describe the important role of the changes in serum M-CSF levels during the menstrual cycle, in the process of fol...
